New Pancreatic Cancer Drug Doubles One-Year Survival Rates in Clinical Trial

An experimental oral medication called daraxonrasib significantly improved survival rates and reduced the risk of death in patients with advanced pancreatic cancer, according to clinical trial data reported by pharmaceutical developer Revolution Medicines and highlighted by major medical institutions. The drug represents a new class of targeted therapies aimed at difficult-to-treat genetic mutations behind one of the deadliest malignancies.

Phase 3 Trial Results and Survival Metrics

According to data released by Revolution Medicines, daraxonrasib reduced the overall risk of death by 60 percent compared to advanced pancreatic cancer patients treated exclusively with standard chemotherapy. Trial participants receiving the medication achieved an average survival rate of 13 months from diagnosis to death, compared to six months for those receiving standard chemotherapy alone, based on a cohort of 168 participants who had previously undergone chemotherapy.

For participants carrying a specific RAS mutation known as G12, tumors remained under control for a median of approximately seven months on daraxonrasib, compared to about three months for patients managed with chemotherapy. Furthermore, approximately 33 percent of patients with the G12 mutation experienced tumor shrinkage or disappearance, compared to roughly 12 percent in the chemotherapy control group. Across the entire trial population, about 31 percent of participants saw their tumors shrink or disappear, versus 11 percent in the chemotherapy arm.

Targeting the RAS Gene With RAS(ON) Inhibitors

Daraxonrasib functions as a first in a new class of drugs called RAS(ON) inhibitors, a novel category of medications designed to target specific variants of the RAS gene that drive cancer growth. Pancreatic cancer has the highest mortality rate of all major cancers, largely due to resistance to conventional therapies and the frequency of RAS pathway alterations. Dr. Mark Goldsmith, chief executive officer and chairman of Revolution Medicines, stated that the findings represent a potentially transformative advance that could redefine the treatment landscape for these patients.

Independent oncologists have also expressed strong optimism regarding the findings. Dr. Diane Simeone, director of the UC San Diego Moores Cancer Center and founder and chief scientific advisor of the Pancreatic Cancer Early Detection Consortium, called the development an extraordinarily hopeful moment for a field that has historically offered far too few effective treatment options. Similarly, Dr. Christina Annunziata, senior vice president of Extramural Discovery Science at the American Cancer Society, noted that the trial results are extremely promising because the drug doubled overall survival for patients with relapsed pancreatic cancer.

Dosage, Administration, and Side Effects

During the trial, participants received oral daily doses of daraxonrasib ranging from 10 milligrams to 400 milligrams, with investigators selecting 300 milligrams as the phase 3 dose. Regarding safety, side effects occurred in 96 percent of participants receiving doses of 300 milligrams or less, with approximately one-third of participants experiencing adverse events of grade 3 or higher.

While the frequency of side effects remains notable, Dr. Annunziata observed that the side effects were lower compared to standard chemotherapy typically used in this second-line treatment setting. However, researchers and oncology advocates note that future availability will depend on the cost and level of access to the new medication.
