The GLP-1 receptor agonist semaglutide extends lifespan and slows physiological aging in healthy older mice, according to a peer-reviewed study published in Nature. Researchers at the University of California, Berkeley found that late-life treatment with the diabetes and weight-loss drug mirrors several anti-aging benefits typically achieved through calorie restriction.

Lifespan and Physiological Impact in Older Mice

In the study, led by UC Berkeley professor of metabolic biology and nutrition Danica Chen, scientists administered semaglutide to 20-month-old female mice for three months to observe its effects during advanced age. According to findings published in the paper, “Late-life semaglutide treatment slows ageing and extends lifespan in female mice,” the treated animals exhibited improved muscle and cognitive function compared to an untreated control group.

Gene expression analysis revealed that the drug successfully attenuated multiple hallmarks of natural aging, including elevated inflammation and diminished tissue regenerative capacity. Furthermore, a separate group of mice treated with semaglutide until the end of life demonstrated a median lifespan nearly 100 days longer than untreated counterparts.

“Most chronic diseases are deeply rooted in the aging process. If GLP-1 agonists do indeed slow it down, then a wide range of clinical benefits is exactly what you’d expect to see,” said Rafael de Cabo, a senior investigator at the NIH’s National Institute on Aging (NIA), who authored a commentary on the research.

Comparison with Calorie Restriction

To determine whether the drug’s longevity benefits stemmed simply from reduced food intake, the researchers compared semaglutide-treated mice against a group placed on a 24% calorie-restricted diet that matched the feeding patterns of the drug group over five months. While both interventions maintained physiological stability across numerous measurements, semaglutide-treated mice surpassed baseline levels in exploratory behavior, spatial memory, and blood-sugar maintenance.

The two groups also diverged in metabolic rate, which decreased in the calorie-restricted animals but remained largely unchanged in the mice receiving semaglutide. According to Danica Chen, these differences suggest that GLP-1 medications engage a biological pathway independent of calorie restriction, pointing toward a distinct route for future longevity interventions.

Clinical Outlook and Human Application

Investigators emphasize that these animal findings do not translate directly to immediate human anti-aging treatments. Additional clinical studies, such as post-hoc analyses of trials like the SLIM LIVER trial, remain necessary to establish whether GLP-1 therapies provide similar longevity efficacy in human patients. Future clinical trials may also target healthy aged populations to test broader preventative applications.
