Following the First Round, MLS clubs will enter the 2027 U.S. Open Cup in the Round of 64 in July, according to Major League Soccer. U.S.-based MLS sides will receive hosting priority for their opening match of the competition. The timing of this entry aligns with preparations for the start of the 2027-28 MLS regular season as the league completes its transition to a new summer-to-spring competition calendar. The tournament adheres to a “one club, one entry” policy and does not place a numerical limit on eligible players for the competition roster. For the 2027 tournament, U.S. Open Cup game day rosters are expanding from 18 to 20 players, with clubs permitted to select from contracted first-team, MLS NEXT Pro, and academy players.

Following the Round of 64, a live draw will be held to reveal matchups and hosting scenarios for the Round of 32 and Round of 16, according to MLS scheduling details. The Round of 32 is scheduled for September 14-15, while Round of 16 dates remain to be confirmed. The Quarterfinals are set for November 23-24, followed by the Semifinals on December 1 and the Final on December 15, though these dates remain subject to change. MLS clubs have participated in the U.S. Open Cup since the league’s inaugural season in 1996, and an MLS side has won every edition of the tournament since 2000.
