The quest for effective weight loss treatments may have an unlikely ally: the python. Researchers have discovered a metabolite found in the blood of pythons that significantly reduces appetite and promotes weight loss in mice, mimicking the effects of popular drugs like Ozempic and Wegovy, but potentially without some of their associated side effects.

The animal kingdom has long been a source of inspiration for pharmaceutical breakthroughs. Semaglutide, the active ingredient in Ozempic and Wegovy, was originally developed based on a hormone found in the venom of the Gila monster, which regulates blood sugar levels. This history highlights the potential for discovering novel therapeutic compounds by studying the unique physiological adaptations of various species.

The Python’s Metabolic Secret: pTOS

Pythons exhibit an extreme feast-and-famine lifestyle. They can consume prey equivalent to 100% of their body weight in a single meal, then go months without eating. This remarkable ability relies on unique metabolic adaptations. Researchers at Stanford Medicine and the University of Colorado, Boulder, investigated the changes that occur in pythons after a large meal, focusing on metabolites – small molecules produced during metabolism.

Their research, published in Nature Metabolism, identified one metabolite, para-tyramine-O-sulfate (pTOS), that increased a thousandfold in python blood after feeding. PTOS is produced by gut bacteria as they break down the amino acid tyrosine.

How pTOS Impacts Appetite and Weight

Surprisingly, pTOS doesn’t naturally occur in mice or rats, the common models for metabolic research. However, when administered to both obese and lean mice, pTOS significantly reduced food intake and led to weight loss. Importantly, this weight loss occurred without the gastrointestinal issues, muscle loss, or energy depletion often associated with other weight loss interventions.

The mechanism behind pTOS’s effects appears to involve the activation of neurons in the ventromedial hypothalamus, a brain region crucial for regulating appetite, hunger, and energy balance. This suggests that pTOS signals to the brain that the body has sufficient energy stores, reducing the drive to eat.

Potential Advantages Over Existing Weight Loss Drugs

Current GLP-1 agonist drugs, like Ozempic and Wegovy, have proven effective for weight loss but can cause side effects such as nausea, vomiting, and diarrhea. Researchers believe pTOS may offer a different approach, potentially minimizing these adverse effects.

Future Directions and Challenges

While the findings are promising, translating pTOS into a human weight loss drug is still a long way off. Further research is needed to understand how pTOS behaves in the human body, determine optimal dosages, and assess its long-term safety and efficacy. Researchers also plan to investigate other metabolites that spike in pythons after a meal, as there are over 200 such compounds.

“We’ve basically discovered an appetite suppressant that works in mice without some of the side-effects that GLP-1 drugs have,” says Leslie Leinwand of the University of Colorado Boulder.
