Wolbachia Mosquitoes: A New Weapon in the Fight Against Dengue

A large-scale trial in Singapore has demonstrated the effectiveness of releasing male Aedes aegypti mosquitoes infected with Wolbachia bacteria in suppressing dengue transmission. The innovative approach, detailed in The New England Journal of Medicine, resulted in a 72% reduction in dengue cases in treated areas.

How Wolbachia Works

Dengue fever, a mosquito-borne viral infection, affects millions globally, causing a spectrum of illness from mild flu-like symptoms to severe complications. Traditional mosquito control methods, such as eliminating breeding sites and using chemical insecticides, often provide only temporary relief and can have negative environmental consequences. The Wolbachia method offers a promising alternative.

Wolbachia is a naturally occurring bacterium found in many insects, but not typically in Aedes aegypti mosquitoes. Researchers from Project Wolbachia–Singapore introduced Wolbachia into male Aedes aegypti mosquitoes. These male mosquitoes, which do not bite humans, are then released into the environment. When they mate with wild female mosquitoes lacking Wolbachia, the resulting eggs do not hatch, leading to a gradual decline in the mosquito population. the National Environment Agency (NEA) of Singapore highlights that Wolbachia can likewise prevent the dengue virus from multiplying within mosquitoes, further reducing their ability to spread the disease.

The Singapore Trial: A Randomized Controlled Study

The study, conducted from mid-2022 to the end of 2024, involved 15 residential areas in Singapore. Eight areas received regular releases of Wolbachia-infected male mosquitoes twice a week, while the remaining seven areas served as a control group. Researchers monitored wild mosquito populations by trapping and counting female mosquitoes and analyzed national health databases to track dengue cases.

The results were significant. Areas with mosquito releases experienced a 77% decrease in the number of wild female mosquitoes – from 0.18 to 0.041 mosquitoes per trap. More importantly, this reduction translated into a substantial decrease in dengue infections. After six months or more, only 6% of residents in treated areas tested positive for dengue, compared to 21% in the control areas. The study demonstrated a 71-72% reduction in dengue risk with exposure to Wolbachia-carrying mosquitoes over a period of three to twelve months .

A Complementary Approach to Dengue Control

The Wolbachia method, also known as Wolbachia-mediated incompatible insect technique combined with sterile insect technique (IIT-SIT), is not intended to replace existing dengue control strategies. Instead, it is envisioned as a complementary tool alongside traditional methods like source reduction and, potentially, vaccines. Experts believe this integrated approach will be most effective in controlling the spread of dengue and other Aedes mosquito-borne diseases.
