New RSV Vaccine Highly Effective in Protecting Older Adults
A new vaccine against respiratory syncytial virus (RSV) has demonstrated significant efficacy in preventing lower respiratory tract disease in adults aged 60 years and older, offering a promising advancement in protecting a vulnerable population. Understanding RSV and Its Impact on Older Adults
Respiratory syncytial virus (RSV) is a common respiratory virus that typically causes mild, cold-like symptoms. Yet, in older adults, RSV can lead to severe illness, including pneumonia, bronchitis, and hospitalization. It is a significant cause of acute respiratory infection, lower respiratory tract disease, and even death in this age group. Currently, no licensed vaccine specifically targets RSV infection, making the development of these new vaccines a crucial step forward in public health.
Clinical Trial Results: Demonstrating Vaccine Efficacy
Recent phase 3 clinical trials have yielded highly encouraging results. A study published in the New England Journal of Medicine evaluated the efficacy and safety of an mRNA-based RSV vaccine, mRNA-1345. The trial involved 35,541 participants over 60 years of age, randomly assigned to receive either the vaccine or a placebo.
The results showed vaccine efficacy of 83.7% (95.88% confidence interval [CI], 66.0 to 92.2) against RSV-associated lower respiratory tract disease with at least two signs or symptoms, and 82.4% (96% CI) against RSV-associated lower respiratory tract disease with at least three signs or symptoms. The vaccine demonstrated 94.1% efficacy (95% CI, 62.4 to 99.9) against severe RSV-related lower respiratory tract disease and 71.7% (95% CI, 56.2 to 82.3) against RSV-related acute respiratory infection. Another study, published in PubMed, assessed the AS01E-adjuvanted RSV prefusion F protein-based candidate vaccine (RSVPreF3 OA). This trial, involving 24,966 participants, showed 82.6% vaccine efficacy (96.95% confidence interval [CI], 57.9 to 94.1) against RT-PCR-confirmed RSV-related lower respiratory tract disease. [2]
Safety Profile of the RSV Vaccines
Importantly, both vaccines demonstrated a favorable safety profile in clinical trials. The mRNA-1345 vaccine showed no evident safety concerns. [3] The RSVPreF3 OA vaccine similarly had an acceptable safety profile. [2] A phase 1 clinical trial of a similar vaccine did not reveal any safety concerns and showed promising immunogenicity in both younger and older adults. [4]
