Protein Linked to Dementia and ALS Also Plays Role in DNA Repair, Raising Implications for Cancer Treatment

A protein associated with neurodegenerative diseases like dementia and amyotrophic lateral sclerosis (ALS) has been found to play a critical role in DNA repair, potentially reshaping our understanding of these conditions and opening new avenues for treatment, including cancer. Researchers at Houston Methodist have discovered that the protein, TDP43, regulates genes responsible for correcting errors that occur when cells copy genetic material – a process known as DNA mismatch repair.

TDP43: A Key Regulator of DNA Repair

The study, published in Nucleic Acids Research, reveals that TDP43 isn’t simply involved in RNA splicing, but is a crucial regulator of the mismatch repair machinery. “DNA repair is one of the most fundamental processes in biology,” said Muralidhar L. Hegde, Ph.D., professor of neurosurgery at the Houston Methodist Research Institute’s Center for Neuroregeneration. “What we found is that TDP43 is not just another RNA-binding protein involved in splicing, but a critical regulator of mismatch repair machinery. That has major implications for diseases like ALS and frontotemporal dementia (FTD) where this protein goes awry.”

The Link to Cancer

Researchers also found evidence connecting TDP43 to cancer. Analysis of large cancer databases showed a correlation between higher amounts of TDP43 and a greater number of mutations in tumors. “This tells us that the biology of this protein is broader than just ALS or FTD,” Hegde explained. “In cancers, this protein appears to be upregulated and linked to increased mutation load. That puts it at the intersection of two of the most vital disease categories of our time: neurodegeneration and cancer.”

Potential Therapeutic Strategies

The findings suggest potential new treatment approaches. In laboratory models, reducing the excessive DNA repair activity caused by abnormal TDP43 partially reversed cellular damage. Controlling DNA mismatch repair may offer a therapeutic strategy for both neurodegenerative diseases and cancer.
