A “forgotten” immune organ, the thymus, may hold the key to better cancer outcomes and lower disease risk. Researchers are reshaping our understanding of the immune system, suggesting this small organ, once believed to become inactive in adulthood, continues to play a vital role throughout life.
What is the Thymus and Why is it Important?
The thymus, located behind the breastbone, is responsible for maturing T cells – critical components of the immune system that fight off infections and disease. For years, it was believed that the thymus shrank with age and eventually stopped functioning, ceasing to produce new T cells in early adulthood. Although, recent research challenges this long-held assumption.
New Research from Aarhus University
Two new studies published in Nature by researchers at Aarhus University in Denmark reveal that a healthy thymus is crucial for both disease risk and the effectiveness of cancer treatments. Professor Nicolai Birkbak, leading the research, explains, “This is significant and potentially paradigm-shifting new knowledge. It challenges the prevailing view that the thymus has no importance in adult life. On the contrary, our studies show that a healthy thymus may be crucial both for disease risk and for how well you respond to cancer treatments.”
Thymus Health and Immunotherapy Outcomes
One study focused on cancer patients undergoing immunotherapy, a treatment that activates the body’s own T cells to attack tumors. The results demonstrated a strong correlation between thymus health and treatment success. Patients with better-preserved thymus function experienced more effective responses to immunotherapy and had longer survival rates. Researchers used a deep-learning framework applied to routine computed tomography (CT) images to quantify thymic health in a cohort of 3,476 patients receiving immune checkpoint inhibitors.
“Our study shows a marked difference in how patients respond to immunotherapy – those with well-preserved thymus function respond better and live longer,” researchers noted.
Lifestyle Factors and Thymus Decline
The research also highlighted the impact of lifestyle on thymus health. Factors such as smoking, obesity, and low physical activity were found to accelerate the shrinkage of the thymus, potentially limiting the body’s ability to produce new T cells. A less functional thymus is associated with an increased risk of early death and the development of cancer or cardiovascular disease.
“Our studies show significant differences in the rate at which the thymus shrinks – both between sexes, but especially in relation to lifestyle factors such as smoking, obesity, and low levels of physical activity,” explains Birkbak.
Future Implications and Potential Interventions
These findings open new avenues for prevention and early intervention. Researchers suggest identifying individuals with rapidly declining thymus function and exploring ways to slow this process could reduce the risk of serious diseases like cardiovascular disease and cancer. This could also potentially improve a patient’s ability to respond to treatment if they are diagnosed with cancer.
“It opens new perspectives. In the long term, it may become relevant to identify individuals with rapid thymus decline and investigate whether it is possible to slow or influence the process, thereby reducing the risk of developing serious diseases such as cardiovascular disease or cancer. This could potentially improve their ability to receive treatment if they are affected,” Birkbak stated.
